Indication of Lanso D 30mg Capsule
  • Acidity occurs when the protective mucous layer of the stomach is eroded, leading to complications such as acidity, GERD (gastroesophageal reflux disease), heartburn, peptic ulcers, and corrosive esophagitis.
  • GERD is characterized by frequent backflow of stomach acid into the food pipe (esophagus), causing irritation and heartburn. Factors contributing to GERD include chocolate, caffeine-containing foods, spicy and fried foods, pregnancy or obesity, lying down after meals, and alcohol or smoking.
  • Corrosive esophagitis occurs when the esophagus is damaged by harmful substances. This can result in ulcers, inflammation, or scarring. Causes of corrosive esophagitis include swallowing strong chemicals like dishwashing liquid, detergents, and drain cleaners, radiation therapy, and certain medications such as painkillers and drugs for osteoporosis.

Lanso D 30mg Capsule relieves symptoms of acid reflux and heartburn. This capsule is not recommended for children below 12 years of age.

Pharmacology

Dexlansoprazole delayed-release capsule is a proton pump inhibitor (PPI) that works by inhibiting the (H+/K+)-ATPase in the gastric parietal cell, leading to a suppression of gastric acid output. It specifically targets the final step of acid generation by acting on the proton pump.

Dexlansoprazole is the R-enantiomer of lansoprazole, which is a racemic mixture of the R- and S-enantiomers. It is available in capsule form with a Dual Delayed Release (DDR) formulation for oral consumption. The capsule contains a combination of two types of enteric-coated granules with different pH-dependent dissolving characteristics.

When taken orally, dexlansoprazole exhibits a plasma concentration-time profile with two separate peaks. The first peak occurs 1 to 2 hours after ingestion, followed by a second peak within 4 to 5 hours. The dosage of dexlansoprazole is associated with proportional increases in the maximum concentration (Cmax) and area under the curve (AUC) values.

Dexlansoprazole is extensively metabolized in the liver and excreted primarily in the urine. The pharmacokinetics of dexlansoprazole may be influenced by factors such as liver function and renal clearance.

It's important to note that the specific dosing regimen and treatment duration of dexlansoprazole should be determined by a healthcare professional based on the individual's condition and response to treatment.

Dosage & Administration of Lanso D 30mg Capsule
  • Lanso D 30mg Capsule is typically used to treat erosive esophagitis and gastroesophageal reflux disease (GERD). The recommended adult dosage for healing erosive esophagitis is 60 mg taken orally once daily for up to 8 weeks. For maintaining the healing of erosive esophagitis or treating heartburn associated with symptomatic non-erosive GERD, the recommended dosage is 30 mg taken orally once daily for up to 6 months or 4 weeks, respectively.
  • For patients with mild hepatic impairment (Child-Pugh A), no dose adjustment is necessary. For those with moderate hepatic impairment (Child-Pugh B), the dosage should not exceed 30 mg per day. Lanso D 30mg Capsule is not recommended for patients with severe hepatic impairment (Child-Pugh C).

The safety and efficacy of Lanso D 30mg Capsules in children below 12 years of age have not been established. For children aged 12 years and above, the recommended dosages for healing erosive esophagitis and treating symptomatic non-erosive GERD are the same as for adults.

To take Lanso D 30mg Capsule, swallow it whole with water. Do not crush or chew the capsule. It is advised to take the medication 1 hour before a meal, preferably in the morning.

Interaction
  • Lanso D 30mg Capsule may interact with various medications and conditions. Some notable drug interactions include painkillers (aspirin, naproxen), anti-depressants (duloxetine, amitriptyline), blood thinners (clopidogrel), hypothyroidism medication (levothyroxine), anti-HIV medication (atazanavir), anti-fungal medication (ketoconazole, itraconazole), anti-cancer medication (methotrexate), vitamin (cyanocobalamin), and sleep aids or relaxants (diazepam, zolpidem). It is important to inform your doctor about all the medications you are taking to avoid potential interactions.
  • Regarding food interactions, it is advised to avoid smoking and alcohol consumption while taking Lanso D 30mg Capsules. Alcohol can increase stomach acid production, leading to increased acidity and heartburn.
  • Lanso D 30mg Capsule may also have interactions with certain diseases or conditions, including Clostridium difficile-associated diarrhea, bone fractures, severe liver disease, low bone mineral density (osteopenia), and low levels of magnesium in the blood. It is essential to discuss your medical history and any existing conditions with your doctor before starting Lanso D 30mg Capsule.
Contraindications
  • If you are allergic to the active component dexlansoprazole, or any other ingredient in the capsule.
  • If you are on or consuming medications that contain Rilpivirine Atazanavir or other anti-HIV medications.
  • If you are consuming herbal products like St. John's Wort (a flowering plant used to treat mood and nervous system-related disorders).
  • If you are on methotrexate (a medicine used in the treatment of cancer and psoriasis-like skin condition).
  • If you are on a blood-thinning agent like warfarin.
  • If you have very low levels of vitamin B-12.
Pregnancy & Lactation
  • The use of dexlansoprazole during pregnancy has not been extensively studied, but observational studies with lansoprazole, the racemic mixture of which dexlansoprazole is the R-enantiomer, did not show an association with adverse pregnancy-related outcomes. Animal studies with lansoprazole showed reductions in offspring femur weight, length, crown-rump length, and growth plate thickness, along with reduced body weight gain. Pregnant women should be advised of the potential risk to the fetus.
  • The presence of dexlansoprazole in human milk and its effects on breastfed infants or milk production is not known. However, lansoprazole and its metabolites have been found in rat milk. The decision to breastfeed should consider the developmental and health benefits of breastfeeding, along with the mother's clinical need for therapy and the potential adverse effects on the breastfed child from both the therapy and the underlying maternal condition.

It is important to consult with a healthcare professional for personalized advice regarding the use of dexlansoprazole during pregnancy or while breastfeeding.

Precautions & Warnings
  • It is important for individuals with specific medical conditions to consult their doctor before taking Lanso D 30mg Capsules. • It is not recommended to take Lanso D 30mg capsule during pregnancy or while breastfeeding. The medication may cause dizziness and sleepiness, so it is important to be alert while driving or operating machinery. Lanso D 30mg Capsule should not be given to children as its safety in this population has not been established.
